Methods and materials
Rats were randomly subjected to six groups: sham operation rats (SOR,
n=6); simple 70%PH group (Group A,
n=24); mild,
moderate,
severe PVS following 70%PH (Group B,
C,
D,
n=24 per group); portal vein ligation following 70%PH (PVL,
n=6) group.
A simple 70%PH was performed in group A as described by Higgins; different degrees of PVS were established by ligating portal trunk with parallel placed needles in different gauges after PH; the portal vein trunk were complete occlusion by thread in PVL rats.
The diameter and velocity of portal vein at the prestenotic (PVDpre,
PVVpre) and stenotic site (PVDs, PVVs) were detected by ultrasound on day 1,
3,
7,
14 after surgery(Fig1;2).
The ratio of diameter (DR) and velocity (VR) was calculated respectively as follows: DR= PVDpre/PVDs, VR= PVV s/ PVV pre.
Rats were weighed on day 1,
3,
7,
14 before sacrificed and blood sample was collected for ALT、AST、TBIL and ALB examination at the same time.
Liver regeneration rate (LRR) which reflected the macroscopic generated liver weight was calculated and morphological changes were observed with microscope in HE staining.The expression of PCNA was investigated by immunohistochemical staining and western blotting.
